Plant growth-promoting characterization of zinc-solubilizing bacteria from rice cultivation in Malaysia as a potential biofertilizer

Abstract

This laboratory experiment was designed to isolate zinc-solubilizing bacteria (ZSB) and evaluate them as a potential plant growth-promoter. Mineral salts medium (MSM) with the addition of insoluble zinc was used as a plate observation for zinc solubilization before the ZSB were characterized further for other biochemical properties. Out of 28 bacterial strains isolated from rice soil, ten were found as ZSB. Isolate SR R-10 was the best ZSB as it obtained a significantly highest solubilization efficiency for zinc oxide (558.33%), zinc carbonate (419.33%) and phosphate solubilization (146.67%). Meanwhile, eight ZSB isolates were found to be able in fixing nitrogen, and seven were positive in siderophores production. In terms of phytohormone production, five selected isolates were tested and SR R-10 led the production of indole-3 acetic acid (IAA) as it produced 6.140 µg mL-1­ followed by SR R-2 and SR N-3 with both produced 5.865 and 5.167 µg mL-1, respectively. 16S rRNA Molecular Identification result shows that SR R-10 is identified as Acinetobacter nosocomialis. This bacteria strain is potentially to be used as a biofertilizer to substitute the chemical fertilizer usage thus promote sustainable agricultural practice.
