Abstract

Production of glucoamylase and cellulase using mixed culture of Aspergillus niger TISTR 3254 and Trichoderma reeseiTISTR3081aswellaspotentialofusingcassavabagasseascarbonsourceforenzymesproductionwereinvestigated. Culturemediacomprisingofcassavabagasse(1-3%)ascarbonsourceinsteadofCMCsupplementedwith1%ofselected nitrogensource(ammonium sulphate,ureaor soybean meal) wereused. Experimentsunder submergefermentation conditionwereconductedatroomtemperatureandundershakingspeedof150rpmduring5days. Theresultsshowedthat mixed culture of Aspergillus niger TISTR 3254 and Trichoderma reesei TISTR3081growninculturemediumcontainingof2% cassavabagasse and1% ammonium sulphate exhibitedthehighest enzyme activity. Glucoamylase enzyme activity of7,734±0.84 U ml -1 andcellulolytic enzyme activitiesof5.76±0.09(FPase) and27.96±0.44(CMCase) U ml -1 were obtained.
